SWAP70 was initially isolated as a component of an isotype switch recombination complex (SWAP) from activated B lymphocytes. It translocates from cytoplasm to the nucleus upon B cell activation. SWAP70 interacts with F-actin and RAC, binds to PI(3,4)P2, and regulates membrane ruffling and lamellipodia formation in response to growth factor stimulation.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1-0.5 µg per ml. For immunoprecipitation, the suggested use of this reagent is 10-30 μg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

- Contains a PH domain, which is required for binding of phosphatidylinositol 3,4,5-trisphosphate, a coil-coil domain, and a domain weakly homologous to DH domains. Theoretical molecular weight is 69 kD.

SWAP70 is localized to cytoplasm in resting B-cells, recruited to plasma membrane, and translocated to the nucleus upon B-cell activation.
- Function
- SWAP70 is a guanine nucleotide exchange factor (GEF) which binds phosphatidylinositol trisphosphate and transduces signals from tyrosine kinase receptors to RAC. SWAP70 is required for membrane ruffling and lamellipodia formation during cytoskeletal F-actin rearrangement.
- Interaction
- Interacts with cytoskeletal F-actin and Rac to regulate actin remodeling. Associates with NPM1, NCL, and PARP1 in switch-associated protein complex.
