Whole cell lysates (15 µg protein) from HeLa (lane 1), Jurkat (lane 2) and NIH3T3 (lane 3) cells were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:1000 (0.5 µg /mL) and 1:2500 (0.2 µg /mL) purified anti-Tubulin-α antibody (clone 10D8). Proteins were visualized using chemiluminescence detection by incubating with 1:3000 dilution of HRP goat anti-mouse IgM secondary antibody. 1:2000 dilution of Direct-Blot™ HRP anti-β-actin antibody (clone 2F1-1, Cat. No. 643807) was used as a loading control (lower).

    IHC staining using purified anti-alpha tubulin (clone 10D8) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using 1X Tris-Buffered Saline (final concentration 0.05 M) with Tween-20 (Cat. No. 925501), the tissue was incubated with 10 μg/mL of antibody overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 594 goat anti-mouse IgM antibody (Cat. No. 405326) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scalebar = 50 μM

Description

Tubulin, one of the major constituents of microtubules, is a dimeric protein consisting of an alpha and beta chain. Tubulin is a highly conserved protein that is ubiquitously expressed. Tubulin is a GTP-binding protein that can be modified by phosphorylation and acetylation. Tubulin interacts with a variety of proteins including RAC GTPase activating protein 1, ZAP70, Polo-like kinase, c-Myc, Fyn, and microtubule associated protein 1A. The 10D8 antibody recognizes α-tubulin in all species and is useful for Western blotting, and immunofluorescence staining. The 10D8 antibody can also be used to monitor protein loading in Western blotting.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, suggested working dilution(s): Use 5 µg per 5 ml antibody dilution buffer for each mini-gel. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 2 - 10 µg/mL is suggested. For immunofluorescent staining applications, a concentration range of 1 - 4 µg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
