VAV1 is one of the three members of the VAV family that functions as a guanine exchange factor (GEF) and is exclusively expressed in hematopoietic and trophoblast cells. VAV1 plays a critical role in immune synapse formation through the regulation of actin cytoskeleton organization. Upon TCR activation, VAV1 is recruited to the immunological synapse and interacts via its SH2 and C-terminal SH3 domains with phosphorylated adaptor protein SLP-76. VAV1 gets phosphorylated and undergoes conformational change to expose its DH domain that modulates GEF activity and promotes GDP to GTP exchange of Rho GTPase CDC42. CDC42 activates WASP and leads to the activation of actin polymerization. In addition, VAV1 transduces T cell receptor signaling including NFAT and NF-κB pathways in a GEF independent manner and promotes T cell activation and development.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.
