FGF-19 belongs to the fibroblast growth factor (FGF) family which includes FGF-19, FGF-21 and FGF-23. FGF-19 binds to FGFR with different affinity, especially with high affinity to FGFR4. α-Klotho and β-Klotho serve as co-receptors to facilitate the binding of FGF-19 to FGFRs in a tissue-specific manner. Though the members of FGF-19 subfamily do not bind to heparin with high affinity, heparin, together with β-Klotho, enhances the interaction between FGF-19 and FGFR4. FGF-19 is involved in regulation of several metabolic processes. Expression of FGF-19 and FGFR4 has been reported to be elevated in human hepatocellular carcinoma (HCC). FGF-19 overexpression in mice FGF-19/FGFR4 stimulates hepatocyte proliferation or tumor formation and invasion via FGF-19/FGFR4 interaction. IL-1β can inhibit β-Klotho expression and FGF-19-induced hepatocyte proliferation. FGF-19/FGFR4 also promotes ovarian cancer proliferation and invasion via AKT-MAPK pathway. FGF-19 are overexpressed in colon adenocarcinoma and lung squamous cell carcinoma. The antibody against FGF-19 can inhibit growth of colon tumor xenografts and hepatocellular carcinoma. FGF-19 is able to increase fatty acid oxidation by enhancing expression of Leptin receptor and by reducing expression of ACC2 [acetyl coenzyme A (CoA) carboxylase 2], the rate-limiting enzyme for fatty acid entry into the mitochondria. A bile acid-activated transcriptional factor, the farnesoid X receptor (FXR), positively regulates FGF-19 expression. In turn, FGF-19 controls bile acid biosynthesis by suppressing expression of a rate-limiting enzyme, cholesterol 7 α-hydroxylase (CYP7A1), via JNK-dependent pathway in human hepatocytes and mouse liver. FGF-19 increase glucose uptake in adipocytes in a β-Klotho-dependent manner.

- Molecular Mass
- The 192 amino acid recombinant protein has a predicted molecular mass of approximately 21.43 kD. The DTT-reduced and non-reduced protein migrates at approximately 27 kD by SDS-PAGE. The predicted N-terminal amino acid is Leu.

- > 95%, as determined by Coomassie stained SDS-PAGE.

- 0.22 µm filtered protein solution is in PBS, 10% glycerol, 5% trehalose

- Less than 0.1 EU per µg cytokine as determined by the LAL method.

- 25 µg sizes are bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
- Activity
- When recombinant human FGFR4 (Cat. No. 752504) is immobilized at 5 µg/mL, recombinant human FGF-19 binds in a dose-dependent manner. The ED50 range for this effect is 3 - 21 ng/mL. Human FGF-19 biotinylated antibody and HRP Avidin (Cat. No. 405103) were used to detect the binding.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
- Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
