Description

Member of the RAB family of small GTPases, RAB10 is a key regulator of intracellular membrane trafficking. Specifically, RAB10 plays a crucial role in the transport of proteins and recycling of endosomes to the plasma membrane. Regulation of RAB10 involves cycling between an active GTP-bound state and an inactive GDP-bound state, controlled by guanine nucleotide exchange factors (GEFs) and GTPase-activating proteins (GAPs). RAB10 phosphorylation at threonine 73 is primarily mediated by Leucine-rich repeat kinase 2 (LRRK2) which plays a crucial role in regulating its interactions with various effector proteins. Dysregulation of RAB10 has been implicated in various diseases, including Charcot-Marie-Tooth disease, a hereditary neurological disorder, and certain types of cancer such as astrocytoma. Additionally, alterations in RAB10 function are associated with neurodegenerative diseases. For example, in Parkinson’s disease, mutations on LRRK2 leads to hyperphosphorylation of RAB10, disrupts normal functions and contribute to the pathophysiology of the disease.

Storage & Handling
Reconstitution: For maximum results, quick spin vial prior to opening. The protein can be aliquoted under aseptic conditions and stored at -20°C or colder. Stock solutions can be prepared in 0.22 µm filtered DI-water and acetonitrile (1:1). It is not recommended to solubilize in PBS or Tris-HCl as the protein shows poor solubility in those buffers. It is recommended to keep the material at a concentration of 1 mg/mL as dimerization becomes more pronounced at lower concentrations.

Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. The product is stable up to 1 month at room temperature but leads to dimerization. Avoid repeated freeze/thaw cycles.

Application Notes

A small portion of the protein in the lyophilized sample may be dimerized (< 5%). To remove dimers, we recommend adding DTT (25 µg/mL) or TCEP (29 µg/mL). DTT should not exceed 0.5 mM to avoid peptide degradation and DTT adducts. TCEP should not be exceed 0.2 mM to avoid forming adducts originating from TCEP excess.

Storing the protein at 4°C and freeze-thaw cycles increases the chance of peptide dimerization.

Structure
RAB10 is a small GTPase protein with a molecular weight of approximately 22.5 kDa. Structurally, RAB10 contains four highly conserved GTPase motifs and a C-terminal geranylgeranylation motif, which are crucial for its membrane association.

Function
RAB10 activity is regulated by guanine nucleotide exchange factors (GEFs) and GTPase-activating proteins (GAPs). GEFs, such as DENND4C and RABIF, facilitate the activation of RAB10 by promoting the exchange of GDP for GTP. On the other hand, GAPs accelerate the hydrolysis of GTP to GDP, thereby inactivating RAB10.
Interaction
RAB10 interacts with kinesin motors KIF13A and KIF13B, which are important for the formation of tubular endosomes.

What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.

Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
