Description

Mouse CD99 (mCD99) is a highly O-glycosylated transmembrane protein that is expressed in endothelial cells and various immune cells, including thymocytes, lymphocytes, monocytes, and neutrophils. CD99 is encoded by the pseudoautosomal gene MIC2. This gene is found in the pseudoautosomal region 1 (PAR1) at the end of the short arm of chromosomes X and Y. The mouse CD99 contains 175 amino acid (aa) with a 28 aa signal sequence, a 109 aa extracellular domain (aa 29-137), a 21 aa transmembrane segment (aa 138-158), and a 17 aa cytoplasmic region (aa 159-175). Mouse CD99 shares 45% and 50% aa sequence identity with the human and rat proteins, respectively. As a cellular ligand for the paired immunoglobulin-like type 2 receptors (PILRs), mCD99 is also called paired immunoglobin-like type 2 receptor-ligand (PILR-L). Mouse CD99 binding to mPILRα relies on the recognition of the Sialylated O-linked sugar chains of mouse CD99, widely involved in the regulation of immune responses. In addition, CD99 regulates various crucial biological processes, including cell adhesion, apoptosis, cell differentiation, and especially plays an important role in the transendothelial migration (TEM) of leukocytes, the critical event for inflammation. CD99 not only serves as a robust marker in cancer, but also as a potential therapeutic target for the treatment of cancers. CD99 influences processes and malignancy associated with several tumors. CD99 is frequently overexpressed in Ewing sarcoma (EWS) and specific subtypes of leukemia. The blockade and knockdown of CD99 in EWS tumor cells promote the inhibition of EWS tumor growth and metastasis. CD99 also modulates the expression of heat shock protein 70 (HSP70), thereby promoting natural killer (NK)-cell activity against acute lymphoblastic leukemia cells. In contrast, CD99-forced expression showed a dramatic negative impact on the survival of osteosarcoma cells, likely through the regulation of caveolin-1 and c-Src kinase activity.

Recombinant mouse CD99-Fc Chimera, amino acid sequence containing Ala27-Gly137 (Accession No. Q8VCN6.1) with a linker (GSSR), and a C-terminal hIgG1-Fc (Pro100-Lys330) was expressed in CHO cells.
Molecular Mass
The 346 amino acid recombinant protein has a predicted molecular mass of approximately 36.88 kD. The DTT-reduced and non-reduced protein migrates at approximately 50 kD and 90 kD by SDS-PAGE, respectively. The predicted N-terminal amino acid is Ala.
Purity
> 95%, as determined by Coomassie stained SDS-PAGE.

Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.

Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency. Please conduct an experiment-specific validation to find the optimal ED50 for your system.
