Mouse Secreted and transmembrane protein 1a (mSECTM1a), also called Protein K-12, is a transmembrane glycoprotein. mSECTM1a shares the greatest homology to human SECTM1. The mouse genome contains two SECTM genes (SECTM1a and SECTM1b) in close proximity to the mouse CD7 gene. Mouse SECTM1a contains N-linked glycosylation motif, a transmembrane domain, and a short cytoplasmic sequence. mSECTM1a is predominantly expressed by cells of the myeloid lineage and epithelia. Similar to human SECTM1, mSECTM1 is also an alternative ligand to CD7 receptor. Both SECTM1a and SECTM1b demonstrate strong affinity and competitively bind to CD7. In addition, mSECTM1 co-stimulates T cell proliferation and IL-2 production in an CD7-independent manner. mSECTM1a activates T cell function through the binding to GITR (glucocorticoid-induced tumor necrosis factor receptor-related protein) rather than CD7. mSECTM1a is defined as a novel regulator for self-renewal of stable Tissue-resident macrophages (TRM). This results in stable TRM populations and favors homeostasis after tissue damage caused by acute inflammation. mSECTM1a may also serve as a new therapeutic agent for the treatment of inflammatory diseases.

- Molecular Mass
- The 375 amino acid recombinant protein has a predicted molecular mass of approximately 42.17 kD. The DTT-reduced and non-reduced proteins migrate at 55 -75 kD and 100 – 140 kD by SDS-PAGE, respectively. The predicted N-terminal amino acid is Gln.
- Purity
- > 95%, as determined by Coomassie stained SDS-PAGE.
- Formulation
- 0.22 µm filtered protein solution is in PBS.
- Endotoxin Level
- Less than 0.1 EU per µg cytokine as determined by the LAL method.
- Concentration
- 25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
- Activity
- When recombinant mouse SECTM1a is immobilized at 5 µg/mL, recombinant mouse CD7 binds in a dose-dependent manner with ED50 range of 35 – 210 ng/mL. HRP anti-His Tag Antibody (Cat. No. 652504) was used to detect the binding.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
