Rat CCL20 was initially identified in ischemic brain tissue and called ST38. In experimental autoimmune panencephalomyelitis, CCL20 expression correlates with the onset of inflammation. In experimental autoimmune encephalomyelitis (EAE), CCL20 and CCR6 mRNAs are induced in the CNS of SJL mice; CCL20 is expressed mainly by CNS-infiltrating leukocytes, and it can be expressed by astrocytes as an additional source of CCL20 during disease relapses. CCL20 is involved in the recruitment of leukocytes to sites of injury and inflammation. CCL20 is constitutively expressed in liver, lung, and mucosal epithelial cells (small intestine and colon). CCL20 is expressed at a low basal level, but its expression is amplified by proinflammatory cytokines and toll-like receptor (TLR) agonists. During infections, CCL20 expression is stimulated in epithelial cells, and immature CCR6+ dendritc cells (DCs) are recruited. Thereafter, the CCR6 expression is downregulated during DC maturation and these migrate to the lymphoid node under the expression of CCR7. CCL20 is the only ligand for CCR6. Nevertheless, mouse and human β-defensin-1 and -2, small cationic antimicrobial peptides, bind and activate CCR6. Contrarily, these β-defensins have lower affinities for CCR6 than CCL20.

- The 71 amino acid recombinant protein has a predicted molecular mass of approximately 8.2 kD. The DTT-reduced protein migrates at approximately 8 kD and non-reduced protein at approximately 10 kD by SDS-PAGE. The N-terminal amino acid is Ala.

- >95%, as determined by Coomassie stained SDS-PAGE.
- Formulation
- 0.22 µm filtered protein solution is in PBS.

- Less than 0.01 ng per µg cytokine as determined by the LAL method.

- 10 and 25 µg sizes are bottled at 200 µg/mL.
- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

CCL20 is expressed in activated cells, including monocytes, T cells, endothelial cells, fibroblasts, Keratinocytes, synoviocytes, dendritic cells, pulmonary and intestinal epithelial cells, and dermal microvascular endothelial cells. CCL20 is induced in astrocytes, miroglia, and macrophages during inflammation.
- Function
- CCL20 chemoattracts immature DCs at sites of injury. CCL20 is upregulated by mediators of inflammation such as TNF or LPS. Also, CCL20 is induced by Th17 cytokines (IL-17A, IL-22, and TNFα) in keratinocytes. CCL20 expression is inhibited by IFNγ and IL-4 in synoviocytes.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
