Biotinylated recombinant CD40 (TNFRSF5)-Fc chimera (Cat. No. 777214) (black circles) binds to immobilized recombinant human CD40L (TNFSF5) (Cat. No. 591706). Ultra-LEAF™ purified anti-human CD154 (CD40L) (clone W21057A) (purple squares) inhibits the binding in a dose-dependent manner, whereas the Ultra-LEAF™ purified rat IgG1, κ isotype control (Cat. No. 400432) (gray triangles) does not have effect. This antibody blocks the binding of 0.3 µg/mL Biotinylated human CD40 (TNFRSF5)-Fc chimera to 1 µg/mL immobilized recombinant human CD40L (TNFSF5). ND50 range: 0.024 - 0.15 µg/mL

Description

CD40L is a type II membrane protein that shares 78% amino acid identity with its mouse counterpart. Human CD40L is a polypeptide of 261 amino acids, consisting of 22, 24, and 215 amino acids for the cytoplasmic, transmembrane, and extracellular domains respectively. Soluble forms of CD40L (sCD40L) have been identified, and an 18 kD fragment has the ability to form homotrimers and bind to CD40 to induce biological signals. The 18 kD protein has full activity in B cell proliferation and differentiation assays. Activated platelets express CD40L on their surface and constitute the major source of sCD40L (>95% in blood). Platelet-derived CD40L regulates neutrophil recruitment and lung damage in sepsis, and it has been suggested that MMP9 regulates CD40L shedding in abdominal sepsis. Platelet-associated CD40L has been linked to progression of atherosclerotic disease, and elevated plasma levels of sCD40L in patients seems to be a reliable predictor of cardiovascular events, such as atherosclerotic plaque rupture and acute coronary syndromes. Also, platelet-associated CD40L expression is increased in idiopathic thrombocytopenic purpura patients, and it has been speculated that platelet-associated-CD40L drives the activation of autoreactive B lymphocytes in this disease. In addition, high plasma levels of sCD40L have been connected to HIV-Associated Neurocognitive Disorders (HAND).

Recommended Usage

Each lot of this antibody is quality control tested by blocking the binding of 0.3 µg/mL recombinant biotinylated human CD40 (TNFRSF5)-Fc (Cat. No. 777214) to immobilized 1 µg/mL recombinant human CD40L (TNFSF5) (Cat. No. 591706). ND50 = 0.024 - 0.15 µg/mL.

T cells, basophils, mast cells, activated platelets

Function
CD40L induces proliferation, differentiation, and immunoglobulin class switching in B cells; also prevents apoptosis in B cell. CD40L is induced in T cells after in vitro activation with PHA, PMA plus ionomycin, anti-CD3, anti-CD2, and CD28 engagement in anti-CD3 activation. CD40L is also induced by thrombin in platelets.
Interaction
B cells, neutrophils, follicular dendritic cells, dendritic cells, macrophages, epithelial cells, epithelial carcinoma cells, endothelial cells, smooth muscle cells, fibroblasts, hepatic stellate cells, and glomerular mesangial cells
Ligand/Receptor
CD40

Related FAQs

Do you guarantee that your antibodies are totally pathogen free?

BioLegend does not test for pathogens in-house aside from the GoInVivo™ product line. However, upon request, this can be tested on a custom basis with an outside, independent laboratory.

Does BioLegend test each Ultra-LEAF™ antibody by functional assay?

No, BioLegend does not test Ultra-LEAF™ antibodies by functional assays unless otherwise indicated. Due to the possible complexities and variations of uses of biofunctional antibodies in different assays and because of the large product portfolio, BioLegend does not currently perform functional assays as a routine QC for the antibodies. However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.

Does BioLegend test each Ultra-LEAF™ antibody for potential pathogens?

No, BioLegend does not test for pathogens in-house unless otherwise indicated.  However, we can recommend an outside vendor to perform this testing as needed.

Have you tested this Ultra-LEAF™ antibody for in vivo or in vitro applications?

We don't test our antibodies for in vivo or in vitro applications unless otherwise indicated. Depending on the product, the TDS may describe literature supporting usage of a particular product for bioassay. It may be best to further consult the literature to find clone specific information.
