Whole cell extracts (15 µg total protein) from HeLa cells untreated or treated with the Cell Activation Cocktail (without Brefeldin A) (Cat No. 423301) were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane and then untreated (left blot) or LPP-treated (right blot). The blots were probed with Purified anti-c-Fos Phospho (Ser32) (clone A22009C) for 2 hours. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control. Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426317) was used as a detection agent. Lane M: Molecular weight marker

No. 423301) for 4 hours (bottom panels) were fixed and permeabilized with 2% PFA and ice-cold methanol, and then intracellularly stained with Purified anti-c-Fos Phospho (Ser32) (clone A22009C) (right panels) or Purified mouse IgG2b, κ Isotype Control (Cat. No. 402201) (left panels) followed by Alexa Fluor® 647 Goat anti-Mouse IgG (Cat. No. 405322). Nuclei were counterstained with DAPI (Cat. No. 422801) (blue) and Alexa Fluor® 488 anti-Cytokeratin (clone C-11) (Cat. No. 628608) (green). The images were captured in a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Description

The proto-oncogene c-Fos is the human homolog of the retroviral v-Fos oncogene, and is a member of the Fos family of transcription factors. c-Fos forms heterodimers with proteins of the Jun family of transcription factors to form the Activator Protein-1 complex (AP-1). This complex is involved in critical cellular processes such as inflammation, cell proliferation, differentiation, survival, hypoxia, bone development and angiogenesis which suggests c-Fos is an important therapeutic target for cancer, vascular remodeling, acute inflammation and autoimmune diseases.  In addition, increases in c-Fos expression and activation are associated with neuronal activity induced by psychoactive drugs. Expression of c-Fos is rapid and transitory, and phosphorylation at residues Ser32 and Thr232 by Erk5 increases protein stability and nuclear localization.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.012 - 0.1 µg/mL. For immunocytochemistry, a concentration range of 2.5 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.06 µg per million cells in 100 µL volume. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For ICFC use we recommend cell fixation/permeabilization with True-Phos™ Perm buffer.

For use of this antibody in immunohistochemistry on formalin-fixed paraffinembedded tissues (IHC-P), we recommend antigen retrieval with Citrate buffer (Cat. No. 420902). However, the antibody also performs well with antigen retrieval with Tris-EDTA (Cat. No. 422703).

Antigen Details

Structure
c-Fos is a 380 amino acid length protein. It has a leucine zipper domain that allows it to form heterodimers with members of the Jun family proteins.
Distribution

Non-phosphorylated forms can be found in the endoplasmic reticulum. However, after activation/phosphorylation the protein is translocated to the nucleus.

Function
c-Fos is a subunit of the AP-1 transcription factor. The half-life of the protein is short; however, phosphorylation induces protein stability and translocation to the nucleus.
Interaction
c-Fos interacts with members of the Jun family proteins to form the AP-1 transcription factor.
Ligand/Receptor
As a part of the transcription factor AP-1, it binds to specific DNA sequences to regulate gene transcription
