HeLa cells were fixed with 1% paraformaldehyde (PFA) and blocked with 5% FBS for 30 minutes. Then the cells were incubated with 10 µg/mL of anti-ATG5 (clone 177.19) Spark YG™ 570 at room temperature 3 hours in dark. Flash Phalloidin™ Green 488 (Cat. No. 424201) was used to stain F-actin. Nuclei were counterstained with DAPI. The image was captured with a 60X objective.

Description

Autophagy protein 5 (ATG5) is involved in autophagic vesicle formation. ATG5 forms a complex with autophagy protein 12 (ATG12), which has E3 ubiquitin ligase activity, and is associated with autophagosome vesicle formation. ATG5 detaches from membranes immediately before or after autophagosome formation. ATG5 is also involved in mitochondrial quality control after oxidative damage, the innate anti-viral immune response, and apoptosis.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 5 - 10 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

* Spark YG™ 570 has a maximum excitation of 555 nm and a maximum emission of 570 nm.
