Recombinant human GM-CSF (Black Circles, Cat. No. 572904) induces the proliferation of human erythroleukemia TF-1 cells. Anti-human GM-CSF antibody (Clone BVD2-21C11, Purple Squares) neutralizes the proliferation function of recombinant human GM-CSF at 0.3 ng/mL in TF-1 cells in a dose dependent manner while the isotype control (Cat. No. 400562, Green Triangles) does not have the effect. The ND50 = 0.1 - 0.8 µg/mL.

Description

Granulocyte/macrophage - colony stimulating factor (GM-CSF) is a hematopoietic factor that is produced by activated T cells, B cells, mast cells, macrophages, fibroblasts, and endothelial cells. In addition to supporting colony formation of granulocyte/macrophage progenitors, GM-CSF is a growth factor for erythroid, megakaryocyte, and eosinophil progenitors.

Preparation
The Ultra-LEAF™ (Low Endotoxin, Azide-Free) antibody was purified by affinity chromatography.
Concentration
The antibody is bottled at the concentration indicated on the vial, typically between 2 mg/mL and 3 mg/mL. Older lots may have also been bottled at 1 mg/mL. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
The antibody solution should be stored undiluted between 2°C and 8°C. This Ultra-LEAF™ solution contains no preservative; handle under aseptic conditions.
Application

Neut - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by neutralization of the proliferation induced by recombinant human GM-CSF (0.3 ng/mL) on TF-1 human erythroleukemia cells. ND50 range: 0.1 - 0.8 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Additional reported applications (for the relevant formats) include: ELISA1-4 or ELISPOT3,4 Detection, Neutralization1, immunohistochemical staining of paraformaldehyde-fixed, saponin-treated5,6 and acetone-fixed7 frozen tissue sections, and immunocytochemistry.

Note: For testing human GM-CSF in serum or plasma, BioLegend's ELISA Max™ Sets (Cat. No. 432001 to 432006) are specially developed and recommended.
