Whole cell extracts (15 µg protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to PVDF and probed with 0.5 µg/mL (1:1000 dilution) of Purified anti-Cyclophilin D Antibody, clone W17239A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker. Lysates were loaded in order of descending PPIF expression (Source: Human Protein Atals). -
A-431 cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with Triton X-100 for 10 minutes, and blocked with 5% FBS for 60 minutes. Cells were then intracellularly stained with a 1:500 dilution (1.0 µg/mL) of either Purified rat IgG2a, κ Isotype Ctrl Antibody (Cat. No. 400502 panel A) or Purified anti-Cyclophilin D Antibody, clone W17239A (Panel B) overnight at 4°C, followed by incubation with Alexa Fluor® 488 Goat anti-rat IgG Antibody (Cat. No. 405418) at 2.0 µg/mL and Alexa Fluor® 647 anti-Cytochrome c Antibody (Cat. No. 612310) at 2.0 µg/mL. Nuclei were counter-stained with DAPI, and the image was captured with a 60X objective.

Cyclophilin D is one of the three components of the permeability transition pore (PTP) localized in the mitochondrial matrix. The opening of the PTP is a key event in the initiation of cell death which leads to mitochondrial swelling, outer membrane rupture, and the release of apoptotic regulators such as cytochrome C. Cyclophilin D promotes mitochondrial permeability transition (mPT) in response to increased Ca2+ level. Cyclophilin D deficient mice have shown desensitization to mTP and require twice as much Ca2+ intake for the opening of the PTP compared to wild-type mice.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.25 - 1.0 µg per ml. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
