HeLa cells were fixed with 1% paraformaldehyde (PFA) for ten minutes, permeabilized with 0.5% Triton X-100 for ten minutes, and blocked with 5% FBS for 30 minutes. Then the cells were intracellularly stained with 1 µg/ml of Alexa Fluor® 488 anti-Tubulin-α (clone 10D8) antibody (green) in 5% FBS for two hours in room temperature. Nuclei were counterstained with DAPI (blue). The image was captured with 60X objective.

Description

Tubulin, one of the major constituents of microtubules, is a dimeric protein consisting of an alpha and beta chain. Tubulin is a highly conserved protein that is ubiquitously expressed. Tubulin is a GTP-binding protein that can be modified by phosphorylation and acetylation. Tubulin interacts with a variety of proteins including RAC GTPase activating protein 1, ZAP70, Polo-like kinase, c-Myc, Fyn, and microtubule associated protein 1A. The 10D8 antibody recognizes α-tubulin in all species and is useful for Western blotting, and immunofluorescence staining. The 10D8 antibody can also be used to monitor protein loading in Western blotting.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistrys, a concentration range of 1.0 - 4.0 µg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 488 has a maximum emission of 519 nm when it is excited at 488 nm.
