Western blot purified anti-Ataxin-2 antibody (clone A17203C). Lane 1: Molecular weight marker; Lane 2: 20 µg of mouse brain lysate; Lane 3: 20 µg of rat brain lysate; Lane 4: 10 µg of SH-SY5Y cell lysate. The blots were incubated with 1 µg/mL of clone A17203C or mouse IgG2b overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

IHC staining of purified anti-Ataxin-2 antibody (clone A17203C) on formalin-fixed paraffin-embedded human spinal cord tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 1 µg/mL of the primary antibody overnight at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

Description

Ataxin-2 is a polyglutamine protein involved in several biological processes including regulation of mRNA translation. Ataxin-2 contains 2 domains, Sm1 and Sm2, that are implicated in RNA splicing and protein interactions. In the brain, ataxin-2 is expressed in the hypothalamus, the brain region that is responsible for controlling energy balance and metabolism homeostasis. Mutations in ataxin-2 are associated with the development of spinocerebellar ataxia type 2.  Ataxin-2 mutant protein with intermediate-length polyglutamine expansion (27-33 polyQ) modifies TDP-43 toxicity and is associated with an increased risk of developing amyotrophic lateral sclerosis.
