Total lysates (15µg protein) from HeLa and HeLa ILK CRISPR/Cas9 knockout (KO) cells were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:500 (1µg/ml) purified anti-ILK antibody , clone P83A9 (upper). Proteins were visualized using chemiluminescence detection by incubation with HRP goat anti-mouse-IgG secondary antibody (cat#405306). Direct-Blot™ HRP anti-β-actin Antibody (Cat#643807) was used as a loading control (lower). Lane M is the Molecular weight ladder. -
Zebra fish tissue lysate and total cell lysates (15µg protein) from SH-SY5Y, HeLa and NIH3T3 were resolved by 4-20% Tris-glycine gel electrophoresis, transferred to nitrocellulose, and probed with 0.5 µg/mL purified anti-ILK (clone P83A9) antibody (upper). Proteins were visualized using a goat anti-mouse-IgG secondary antibody conjugated to HRP and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin antibody (middle) and Ponceau S staining (lower) were used as a loading controls. -
HeLa cells were fixed with 4% PFA for fifteen minutes, permeabilized with 0.5% Triton X-100 for three minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with 2 µg/mL of purified anti-ILK antibody (clone P83A9) overnight at 4°C followed by Alexa Fluor® 594 (red) conjugated goat anti-mouse IgG antibody for one hour at room temperature. Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective.

Integrin-linked kinase (ILK) is an integrin associated actin and tubulin cytoskeletal interacting effector. ILK was identified in a search for proteins interacting with integrin β CTs. It has an N-terminal ankyrin repeat domain (ARD) and a C-terminal integrin β CT binding kinase-like domain (KLD). ILK functions as a key scaffold at cell-ECM adhesions and regulates integrin-mediated as well as growth factor regulated functions. ILK orchestrates several signaling pathways, such as PI3Kinase/Akt, Wnt, TGF-β, Hippo tumor suppressor pathway and epithelial-mesenchymal transition signaling in various types of cancer cells. ILK expression is upregulated in many types of cancers.
