    Mouse brain (lane 1), mouse liver (lane 2), N-tera2 (lane 3), and NF-1 (lane 4) cell extracts were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified monoclonal tubulin beta 3 (TUBB3) (clone AA10) antibody. Proteins were visualized using an anti-mouse-IgG secondary conjugated to HRP and chemiluminescence detection.

    Whole cell lysates (15 µg protein) from HEL (negative control), HeLa, NTERA-2, and A549 cells were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, and probed with 0.25 µg/mL (1:2000 dilution) of purified anti-Tubulin Beta 3 (TUBB3) antibody, clone AA10. Proteins were visualized using chemiluminescence detection by incubating with 1:3000 dilution of HRP goat anti-mouse-IgG secondary antibody (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH antibody (1:2000 dilution, Cat. No. 649203) was used as a loading control (lower). Lane M: MW ladder. Cell lysates were loaded in order of increasing TUBB3 mRNA expression levels (source: Human Protein Atlas).

    NTERA‐2 cells were fixed with 4% paraformaldehyde (PFA) for 10 minutes, permeabilized with 0.5% Triton X‐100 for 5 minutes, and blocked with 5% FBS for 30 minutes. Then the cells were intracellularly stained with 2.5 µg/mL anti‐Tubulin Beta 3 (TUBB3) (AA10) antibody and followed by DyLight™ 594 goat anti‐mouse IgG for one hour at room temperature. Nuclei were counterstained with DAPI (blue). The image was captured with a 40X objective.

Description

Tubulin is the main component of microtubules. In adults, tubulin beta 3 (TUBB3) is primarily expressed in neurons and is commonly used as a neuronal marker. It plays an important role in neuronal cell proliferation and differentiation. Mutations in this gene cause congenital fibrosis of the type 3 extraocular muscles. Tubulin beta 3 (TUBB3) is also found in a wide range of tumors. Studies indicate that it is a predictive and prognostic marker in various tumors.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.025 - 0.25 µg per ml. For immunocytochemistry, the suggested use of this reagent is 1.0 - 5.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
