Total cell lysates (15 µg protein) from 293T (negative control), A431 and NIH3T3 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to nitrocellulose, and probed with 2 µg/mL purified anti-Claudin-1 (clone 9E12C72) antibody, competitor’s antibody used at manufactures recommended concentration (upper). Proteins were visualized by chemiluminescence detection using a goat anti-mouse-IgG secondary antibody conjugated to HRP for anti-Claudin-1 antibody, and a donkey anti-rabbit IgG Antibody conjugated to HRP for competitor’s antibody. Direct-Blot™ HRP anti-β-actin Antibody was used as a loading control (lower). Lane M: molecular weight ladder and M* indicates extended exposure of the molecular weight ladder.

Description

Claudin-1 is a member of the claudin family of proteins, which is composed of at least 24 members. Claudin proteins are integral membrane proteins with 4 transmembrane regions and 2 extracellular loops that form tight junctions. Tight junctions function as a channel and a barrier to maintain cellular morphology, polarity, and homeostasis between epithelial cells. Claudin-1 is a main component of tight junctions and is sorely responsible for normal barrier function of the skin. Misregulated expression of Claudin-1 has been shown to be associated with cancer progression and metastasis in several cancers. Claudin-1 has been found to be overexpressed in neoplasia associated with inflammatory bowel disease. Mutations in Claudin-1 gene lead to human skin diseases and Claudin-1 deficient mice die soon after birth due to dehydration.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

This clone has no cross-reactivity with mouse (in house tested).
