C57BL/6 peritoneal macrophages were first treated with TruStain fcX™ (anti-mouse CD16/32) Antibody and True-Stain Monocyte Blocker™. Cells were then stained with PE anti-mouse CD209a (clone MMD3) (above) or PE mouse IgG2a, κ isotype control (below) and F4/80 (clone BM8) Alexa Fluor® 647. Data shown was derived from CD3/CD19/NK-1.1 negative populations.

Description

CD209a, is also known as dendritic cell-specific intercellular adhesion molecule 3-grabbing nonintegrin (DC-SIGN) and CIRE. Of the eight DC-SIGN-related receptors in mice, CD209a is considered the closest homolog to human DC-SIGN (CD209). Although both mouse and human counterparts are expressed on monocyte-derived dendritic cells, expression variation has been noted. Mouse CD209a is found on skin and splenic myeloid cells and is highly expressed on plasmacytoid dendritic cells, while human CD209 is expressed on myeloid dendritic cells, placental macrophages, liver and placental endothelial cells. CD209 binds to ICAM-3 (CD50), ICAM-2 (CD102), and Butyrophilin (BTN2A1), and mediates dendritic cell migration and T cell proliferation. It plays a critical role in capturing and internalizing pathogens such as Streptococcus pneumoniae.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 μg per million cells in 100 µl volume. It is recommended that the reagent be titrated for optimal performance for each application.
