    C57BL/6 mouse frozen spleen section was fixed with 4% paraformaldehyde (PFA) for 10 minutes at room temperature and blocked with 5% FBS for 30 minutes at room temperature. Then the section was stained with 10 µg/ml of CD73 (clone TY/11.8) Brilliant Violet 421™ (blue), CD3 (clone 17A2) Alexa Fluor® 594 (red), and B220 (clone RA3-6B2) Alexa Fluor® 647 (green) overnight at 4°C. The image was captured by 10X objective.

Description

CD73 (ecto-5'-nucleotidase) is a 69 kD GPI-anchored surface protein. In mice, expression of CD73 in bone marrow is restricted to CD11b+ myeloid cells. In spleen, it is largely expressed on T cells.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤0.125 µg per million cells in 100 µl volume. For immunohistochemical staining on frozen tissue sections, the suggested use of this reagent is 5.0 - 10.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Brilliant Violet 421™ excites at 405 nm and emits at 421 nm. The standard bandpass filter 450/50 nm is recommended for detection. Brilliant Violet 421™ is a trademark of Sirigen Group Ltd.
