IHC staining of purified anti-Myelin Protein Zero antibody (clone 58A) on formalin-fixed paraffin-embedded mouse sciatic nerve tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 5 µg/ml of the primary antibody overnight at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. Western blot of purified anti-Myelin Protein Zero antibody (clone 58A). Lane 1: Molecular weight marker; Lane 2: 20µg of mouse sciatic nerve lysate; Lane 3: 20µg of rat sciatic nerve lysate. The blot was incubated with 1 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

Myelin protein zero (MPZ), is a membrane glycoprotein mainly expressed in the peripheral nervous system (PNS). It is the major structural component of the myelin sheath, and accounts for at least 50% of all proteins in the PNS. MPZ is expressed by Schewann cells. Mutations in MPZ have been associated with myelin deficiency, which may lead to neuropathies including charcot-Marie-Tooth and Dejerine-Sottas diseases.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5 - 10 µg/mL is suggested. For western blotting, the suggested use of this reagent is 1 - 5 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
