Following antigen retrieval using Sodium Citrate H.I.E.R., 1X (Cat. No. 928502), the tissue was incubated 5 μg/mL of purified anti-GFAP (Cat. No. 644702) followed by incubation with 10 μg/mL of Spark Red™ 718 Goat anti-mouse IgG for 1 hour at room temperature. Nuclei were counterstained with DAPI (blue, Cat. No. 422801).     HeLa cells were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 5 μg/mL of purified anti-Cofilin 1 (Cat. No. 865302), followed by incubation with 10 μg/mL Spark Red™ 718 Goat anti-mouse IgG (clone Poly4053) (orange, panel A). Cells were also stained with Flash Phalloidin™ Green 488 (Cat. No. 424201) for 1 hour at room temperature and nuclei were counterstained with DAPI (Cat. No. 422801) (merge in panel B). The images were captured by a Revvity Operetta CLS™ High Content Analysis System with a 60X objective. Scale bar: 50 μm

Description

This polyclonal goat anti-mouse IgG antibody reacts with the heavy chains of mouse IgG and with the light (kappa and lambda) chains common to most mouse immunoglobulins. No cross-reactivity has been detected against non-immunoglobulin serum proteins. This antibody has been solid-phase absorbed to ensure minimal cross-reaction with rat, human, bovine, horse, and rabbit immunoglobulins, but it may have minimal cross-reactivity with other subclasses of mouse immunoglobulins.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5.0 - 10.0 µg/mL is suggested. For immunohistochemistry on frozen tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 2.5 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

* Spark Red™ 718 has a maximum excitation of 697 nm and a maximum emission of 718 nm.
