C57BL/6 mouse splenocytes were stained with purified CD11a (clone 2D7) (filled histogram) or rat IgG2a, κ isotype control (open histogram) followed by anti-rat IgG FITC antibody.

    C57BL/6 mouse frozen spleen section was fixed with 4% paraformaldehyde (PFA) for 10 minutes at room temperature and blocked with 5% FBS for 30 minutes at room temperature. Then the section was stained with 10 µg/mL purified anti-mouse CD11a (clone 2D7) and anti-mouse CD3ε (clone 500A2) Alexa Fluor® 594 (green) overnight at 4°C, followed by 2.5 µg/mL of Alexa Fluor® 647 anti-rat IgG (Poly4054) (red) for two hours at room temperature. Nuclei were counterstained with DAPI (blue). The image was captured by 10X objective.

Description

CD11a is a 180 kD glycoprotein, also known as αL integrin, LFA-1 α, Ly-15, or Ly-21. It is a member of the integrin family, primarily expressed on lymphocytes, monocytes/macrophages, and granulocytes. In association with CD18, the CD11a/CD18 complex forms LFA-1. CD11a plays an important role in intercellular adhesion and costimulation by binding its ligands, ICAM-1 (CD54), ICAM-2 (CD102), and ICAM-3 (CD50).

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. For immunohistochemistry on frozen tissue sections, a concentration range of 5.0 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
