TOX is a member of the high-mobility group (HMG) box superfamily of DNA-binding proteins and is originally identified as the thymic transcript expressed in CD4+CD8+ double positive T-cells. TOX is also defined as part of a small subfamily of transcription factors consisting of TOX2, TOX3, and TOX4, which share conserved HMG-box sequences. TOX is highly upregulated in thymocytes undergoing positive selection and is required for the differentiation and development of various T-cell subsets, such as CD4+ T cells, regulatory T cells, and natural killer T cells (NKTs). TOX-deficient mice fail to develop lymph nodes and have significantly diminished numbers of CD4+ T-cells, NK cells, and ILC cells.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 µg per ml. For immunocytochemistry, a concentration range of 0.1 - 10 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

The immunogen used to generate clone 6E6D03 is the N-terminus of Tox1, which is highly conserved among Tox family member proteins. Hence, this clone may recognize multiple Tox family members. Predicted MW for Tox family members: Tox 1 is 57.5 kD, Tox 2 is 49, 52, 53, 55 kD, Tox 3 is 63 kD and Tox 4 is 64, 66 kD.
