Total cell lysates (15 µg total protein) from E. Coli BL21 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 0.01 µg/mL (1:100,000 dilution) of Purified anti-E. Coli RNA Polymerase β Antibody, clone 8RB13, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Lane M: Molecular Weight marker.

Description

RNA polymerase (RNAP or RNApol) is an enzyme that produces primary transcript RNA. The β subunit is the second-largest subunit, and is encoded by the roB gene. RNA polymerase β is a subunit which, along wtih α (rpoC), limits core polymerase expression.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. To reconstitute, add 100 µl of ddH2O to the tube containing antibody and mix to dissolve. For Western blotting, the suggested dilution is 1:1000 after reconsitution. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone 8RB13 is extremely useful for detecting or purifying core RNAP from a broad range of bacteria. The purified polymerase is highly active for in vitro transcription.

Additional reported applications (for the relevant formats) include: immunoprecipitation3. The epitope of clone 8RB13 has been mapped to the sequence PEEKLLRAIFGEKAS3.
